  • Retroviral integrase (IN) is an enzyme produced by a retrovirus (such as HIV) that integrates (forms covalent links between) its genetic information into that of the host cell it infects.
  • Temperate bacteriophages integrate into the host genome at specific interrupted dyad symmetry sequences using the phage encoded enzyme integrase (see prophage integration).
  • The reverse transcriptase and integrase proteins are needed for the retrotransposon activity of the virus.
  • Its innermost region consists of a cone-shaped core that includes two copies of the (positive sense) ssRNA genome, the enzymes reverse transcriptase, integrase and protease, some minor proteins, and the major core protein.
  • The enzyme (38 kDa) is a member of the integrase family of site specific recombinase and it is known to catalyse the site specific recombination event between two DNA recognition sites (LoxP sites).
  • The integron consists of a promoter, an attachment site, and an integrase gene that encodes a site-specific recombinase There are three classes of integrons described.
  • Notably, retroviral genomes are composed of single-stranded RNA and must be converted to proviral double-stranded DNA, a process known as reverse transcription—before it is integrated into the host genome via viral proteins like integrase.
  • p21 mediates the resistance of hematopoietic cells to an infection with HIV by complexing with the HIV integrase and thereby aborting chromosomal integration of the provirus.
  • As an integrase inhibitor, raltegravir targets integrase, an enzyme common to retroviruses that integrates the viral genetic material into human chromosomes, a critical step in the HIV infection model.
  • Typically, an element flanked by a pair of LTRs will encode a reverse transcriptase and an integrase, allowing the element to be copied and inserted at a different location of the genome.
  • Elvitegravir is a low-molecular-weight, highly selective integrase inhibitor that shares a core structure with quinolone antibiotics.
  • The earliest identified members of the serine recombinase family were known as resolvases or DNA invertases, while the founding member of the tyrosine recombinases, lambda phage integrase (using attP/B recognition sites), differs from the now well-known enzymes such as Cre (from the P1 phage) and FLP (from the yeast Saccharomyces cerevisiae).
  • LEDGF/p75, a transcription coactivator, gained prominence as a host factor that assists HIV integration and is probably the only integrase interactor whose knock-down severely affects the HIV integration levels.
  • In the European Union, rilpivirine is approved in combination with cabotegravir for maintenance treatment of adults who have undetectable HIV levels in the blood (viral load less than 50 copies/ml) with their current antiretroviral treatment, and when the virus has not developed resistance to certain class of anti-HIV medicines called non-nucleoside reverse transcriptase inhibitors (NNRTIs) and integrase strand transfer inhibitors (INIs).
  • The function of the N-terminal domain is not completely clear, but is thought to assist the integrase in forming multimers (fixed agglomerations of multiple integrase molecules).
  • These include Januvia (the first DPP-4 inhibitor for type 2 diabetes), Gardasil (the first vaccine for prevention of cervical cancer), Isentress (the first HIV integrase inhibitor), Zostavax (the first vaccine for the prevention of shingles in adults), and Rotateq (an oral vaccine for the prevention of rotavirus infection in infants).
  • Chicoric acid has been shown to stimulate phagocytosis in both in vitro and in vivo studies, to inhibit the function of hyaluronidase (an enzyme that breaks down hyaluronic acid in the human body), to protect collagen from damage due to free radicals, and to inhibit the function of HIV-1 integrase.
  • Retroviral aspartyl protease is synthesised as part of the pol polyprotein that contains an aspartyl protease, a reverse transcriptase, RNase H and integrase.
  • Like ritonavir (Norvir), cobicistat is of interest for its ability to inhibit liver enzymes that metabolize other medications used to treat HIV, notably elvitegravir, an HIV integrase inhibitor.
  • This analysis concluded in the localization of the upper pathway genes of naphthalene degradation, right next to the genes encoding tRNA-Gly and integrase, as well as the identification of the genes encoding enzymes involved in the degradation of salicylate, benzoate, 4-hydroxybenzoate, phenylacetic acid, hydroxyphenyl acetic acid, and the recognition of an operon involved in glucose transport in the strain.
